A highly concentrated analyte in an @A00339@ can be determined with better @P04799@ by replacing the @M03756@ by one containing a solution of the analyte or other @A00034@ of known concentration; this is known as difference absorption @S05848@. Difference spectra can also be obtained by computer or other subtraction methods.
